Tınlı bir toprağın katalaz aktivitesi ve kinetiği üzerine 2,4-D (Diklorofenoksiasetik Asit) herbisitinin etkisi
Bu çalışmada, tın bünyeli toprağa artan düzeylerde (0.5 ppb, 1 ppb ve 2 ppb) 2,4-D herbisit uygulanmasının farklı substrat konsantrasyonları ve zamana bağlı olarak toprakların katalaz enzim aktivitesi değişimleri ve bu enzim reaksiyonunun kinetik parametreleri (Vmax, Km ve Vmax/Km) üzerine etkisi inkübasyon denemesi ile araştırılmıştır. Bu amaçla deneme, tesadüf parselleri deneme desenine göre 3 paralelli olarak OMÜ Ziraat Fakültesi Toprak Bölümü laboratuarında kurulmuş ve yürütülmüştür. Deneme süresince topraklardan eksilen su miktarı her gün tartılarak maksimum su kapasitesinin %40’ı seviyesinde olacak şekilde tamamlanmıştır. Denemenin 15., 30., 45., 60., 75., ve 90. günlerinde alınan toprak örneklerinin katalaz enzim aktiviteleri belirlenerek bu enzime ait kinetik parametreler hesaplanmıştır. Effect of 2,4-D (Dichlorophenoxyacetic Acid) herbicide on catalase activity and kinetic parameters of a loamy soil
In this study, effects of increasing dose application (0.5 ppb, 1 ppb and 2 ppb) of 2,4-D herbicide on changes in catalase enzyme activity and kinetic parameters (Vmax, Km and Vmax/Km) in a loamy soil were investigated depending on substrate concentration and incubation time. For this purpose, an experiment was conducted in a randomized plot design with three replicates and carried out at the laboratory of Soil Science Department of Agricultural Faculty at Ondokuzmayis University. During the experiment, decreased moisture level in soils was completed to 40% of maximum moisture holding capacity by weighing everyday. Catalase enzyme activity was determined in soil samples taken 15., 30., 45., 60., 75. and 90. days of the experiment, and kinetic parameters of this activity were also calculated.
